The present study aimed to use the primary hepatocyte culture of orang spotted grouper, Epinephlus coioides for assessing the toxic effects of nonylphenol (NP) on the primary hepatocyte viability and hepatocyte damage. For this purpose, the liver of eight specimens of E. coioides were dissected and the hepatocytes were enzymatically digested, isolated and counted. The IC50 of the toxicants on cells was determined separately using MTT assay. Then the hepatocytes were exposed to 10, 20 and 30% of IC50 concentrations (M 5-10، M 5-10×2 و M 5-10×3) ). To determine the effects of pollutants, the cultivated hepatocyts were sampled after 0, 6, 12 and 24 h of exposure. According to the results, the lower concentrations of toxicant did not show negative effects on the cell growth, however, the cell damage increased in a dose- and time-dependent manner. In conclusion, this study revealed that NP has a cytotoxic effect on primary hepatocytes.
