Polyhydroxybutyrate is one of the biodegradable compounds which is produced by cyanobacteria Spirulina sp. In this study, cyanobacteria Spirulina sp isolated from water reservoirs of Guilan province were cultured in Zarrouk medium for 20 days and were studied under autotrophic, heterotrophic and mixotrophic conditions. To evaluate the quality of extracted PHB, staining with Sudan Black and FTIR methods were used and then gas chromatography (GC) was used for quantitative analysis. Polyhydroxy butyrate granules in treated Spirulina sp. cells in all three autotrophic, mixototropic and heterotrophic culture medium were observed as black spots. Regarding the FTIR absorption spectrum, the presence of polyhydroxy butyrate isolated from Spirulina sp. was confirmed. The concentration of polyhydroxy butyrate in autotrophic, mixotrophic and heterotrophic conditions according to the GC chromatogram were 0.414, 0.794 and 0.816 μg/l, respectively. According to the results, it seems that the heterotrophic culture medium is more suitable than other two environments for polyhydroxy butyrate accumulation.
